Plant cytokinin ortho-topolin riboside suppresses the proliferation of multiple myeloma cells by promoting β-catenin degradation.

Kim Minji M, Kang Wonku W, Oh Sangtaek S

Multiple myeloma involves the aberrant activation of the Wnt/β-catenin signaling pathway, resulting in β-catenin accumulation. Targeting this pathway presents a promising therapeutic approach for multiple myeloma. A cell-based chemical assay revealed that ortho-topolin riboside (oTR), a cytokinin naturally produced by Populus robusta, effectively suppressed the activation of β-catenin response transcription induced by Wnt3aconditioned medium. The mechanism underlying this suppression involved oTR-triggered phosphorylation of β-catenin at Ser33, Ser37, and Thr41, leading to its proteasomal degradation and a reduction in intracellular β-catenin levels. Furthermore, oTR consistently decreased β-catenin levels and inhibited the expression of Axin2, cyclin D1, and c-Myc-genes regulated by β-catenin/T cell factor signaling in multiple myeloma cells. Additionally, oTR inhibited the proliferation of multiple myeloma cells and promoted apoptosis, as evidenced by an increase in Annexin V-FITC-positive cells and elevated caspase-3/7 activity. These findings suggest that oTR exerts its antiproliferative effects by attenuating the Wnt/β-catenin pathway, indicating its potential as a therapeutic agent for preventing and treating multiple myeloma.

Denitrogenative thiolation of benzotriazinones: facile access to ortho-thiolated benzamides.

Chaithanya Dasari D, Sakthiganapathi Radhakrishnan R, Jhansi Jaladi J, Korivi Ramaraju R et al.

A metal-free denitrogenative thiolation of 1,2,3-benzotriazin-4(3H)-ones with thiols for the synthesis of ortho-thiolated benzamides in the presence of DBU is described. The reaction accommodates a broad range of N-aryl-substituted 1,2,3-benzotriazin-4(3H)-ones, as well as aryl and alkyl thiols, affording the desired products in moderate to high yields. The reaction is believed to proceed via a benzenediazonium intermediate. The scalability of the developed protocol was also further demonstrated.

Regioisomeric engineering of carbazole-naphthalimide donor-acceptor systems enables efficient triplet generation and room temperature phosphorescence in molecular and nanoaggregated states.

Mancini Pietro P, Gouthaman Siddan S, Rajendran Vignesh V, Montanari Chiara C et al.

Regioisomeric engineering provides an effective strategy for controlling excited-state dynamics in donor-acceptor systems, yet its influence on triplet generation and room-temperature phosphorescence (RTP) remains insufficiently understood. Herein, we report two series of carbazole-phenyl-naphthalimide donor-acceptor regioisomers in which carbazole is connected to either the C3 or C4 position of the naphthalimide acceptor through ortho-, meta-, and para-substituted phenyl linkers. Systematic photophysical investigations reveal that subtle changes in molecular connectivity profoundly influence charge-transfer character, fluorescence and intersystem crossing efficiency. Para-linked and C4-substituted derivatives exhibit enhanced fluorescence owing to improved conjugation and reduced structural relaxation, whereas ortho- and meta-isomers as well as C3-substitution promote more efficient triplet generation through spin-orbit charge-transfer-assisted intersystem crossing, yielding triplet quantum yields approaching unity. Femtosecond and nanosecond transient absorption spectroscopy directly elucidate the singlet-to-triplet conversion dynamics, while TD-DFT calculations and single-crystal X-ray diffraction establish clear relationships between molecular geometry, electronic coupling, and excited-state behaviour. Efficient orange-to-red RTP is observed in benzophenone host-guest matrices as well as in water-dispersed nanoaggregates, suggesting that regioisomeric control can activate persistent phosphorescence in both solid-state and colloidal environments. These findings provide valuable molecular design principles for developing purely organic RTP materials for optoelectronics, time-resolved bioimaging, and photonic applications.

Prenatal exposure to di(isononyl) cyclohexane-1,2-dicarboxylate and adverse birth outcomes in the USA: a prospective analysis of births in a large American birth cohort.

Trasande Leonardo L, Nelson Morgan E ME, Alshawabkeh Akram A, Barrett Emily S ES et al.

A large body of evidence has documented the effects of ortho-phthalates, additives to soft plastic used in food packaging, on premature birth. Ortho-phthalates are increasingly replaced with alternatives including tere-phthalates and di(isononyl) cyclohexane-1,2-dicarboxylate (DINCH). A limited toxicological literature suggests that DINCH metabolites disrupt sex hormones and promote oxidant stress known to contribute to adverse birth outcomes, and epidemiologic studies have also been limited in sample size, inclusivity of different populations and measurements at different time points in pregnancy. We leveraged a large American cohort comprised of mother-child dyads from twenty-two cohorts to assess associations of trimester-specific and pregnancy-wide DINCH metabolites with birth weight (BW) and gestational age (GA). Average prenatal urinary DINCH was inversely associated with birth length, birthweight, and birthweight for gestational age z-scores, but not GA. A tenfold increase in the pregnancy-averaged DINCH metabolite cyclohexane-1,2-dicarboxylic acid-monocarboxy isooctyl ester (MCOCH) was associated with a decrease of 27.43 g (95%CI: -54.81, -0.04) in birthweight. Trimester-specific analyses revealed stronger associations for third trimester DINCH exposure with birth outcomes. In a large American cohort representing a broad range of backgrounds, we identified an inverse association of DINCH with standardized measures of birth size, but not gestational age. Further studies should examine joint exposures to DINCH, tere- and orthophthalates, and repeated measures across pregnancy.

Design, synthesis, and pharmacological evaluation of 7-substituted 1,2,4-triazolo[1,5-a]pyrimidine derivatives as novel antiepileptic lead candidates targeting Nav1.2 channels.

He Luqian L, Zhang Qiaoliang Q, Ma Wenjuan W, Qian Meiqi M et al.

The limitations of current antiepileptic drugs, including drug resistance and adverse effects, necessitate the development of novel therapeutic agents with improved efficacy and safety. Herein, we describe the design, synthesis, and pharmacological evaluation of a series of novel 1,2,4-triazolo[1,5-a]pyrimidine derivatives. Through strategic modification at the C-7 position, we identified compound 5m as a highly potent anticonvulsant agent. In the pentylenetetrazole (PTZ) epilepsy model, 5m exhibited exceptional antiseizure activity (ED50 = 8.84 mg/kg) with a high protective index (PI > 14.80), demonstrating superior efficacy and safety profiles compared to valproate, carbamazepine, and phenobarbital. Notably, 5m showed no neurotoxicity at its maximum soluble concentration. Preliminary structure-activity relationship (SAR) analysis at the C-7 position suggested that the trifluoromethanesulfonate substituent, the pyrimidine nitrogen atoms, and the ortho-fluoro substitution pattern on the phenyl ring are important for potent activity. Mechanism of action studies combining electrophysiological patch-clamp recordings and molecular docking simulations established Nav1.2 channels as a primary molecular target, with 5m exhibiting inhibitory activity against Nav1.2 channels (23% ± 17% inhibition at 10 μM) and forming key interactions with residues LEU329, LEU336, ASN388, GLN391, and ARG395, while showing no significant effect on GABAA receptors. Furthermore, in silico ADMET prediction confirmed that 5m possesses excellent drug-like properties, including favorable gastrointestinal absorption and blood-brain barrier permeability, with no predicted toxicity. In summary, compound 5m represents a promising novel antiepileptic lead candidate with high efficacy, low neurotoxicity, and a favorable drug-like profile, warranting further preclinical development.

Positional Isomerism: A Molecular Switch for Substantial Modulation of Polymer Mechanics.

Yang Chengna C, Liu Guoquan G, Bai Ruixue R, Jing Xinli X

Positional isomerism, a universal structural regulation strategy in nature, achieves precise control over biological functions through minute variations in substituent positions on a molecular backbone. Despite its extensive application in small-molecule chemistry, the systematic introduction of positional isomerism as the sole structural variable into polymer main chains remains unexplored. Herein, inspired by biological systems, we employ three isomeric aromatic diols, 1,2-, 1,3-, and 1,4-benzenedimethanol (o-, m-, and p-BDM) as chain extenders to synthesize three thermoplastic polyurethanes with identical chemical composition, differing only in the substitution pattern on the aromatic ring. The para-substituted polyurethane (p-PU) exhibits a tensile strength of 13.1 MPa, an elongation at break of 2016%, and a toughness of 151.8 MJ/m3, representing a 7.7-fold increase in tensile strength and a 19.2-fold increase in toughness over its ortho-substituted counterpart (o-PU, with a tensile strength of 1.7 MPa, an elongation at break of 604%, and a toughness of 7.9 MJ/m3), while the meta-substituted polyurethane (m-PU) shows intermediate values (tensile strength: 7.4 MPa; elongation at break: 1271%; toughness: 47.0 MJ/m3). Combined characterization using small-angle X-ray scattering (SAXS), density functional theory (DFT) calculations, and molecular dynamics simulations reveals that the para isomer, with minimal steric hindrance, promotes ordered hydrogen bonding and π-π stacking within hard segments, inducing the most well-defined microphase-separated morphology among the three isomers. This work demonstrates the feasibility of positional isomerism as an independent dimension for tuning polymer properties and offers a molecular design strategy for precisely modulating mechanical performance without altering chemical composition.
