A subunit vaccine protects A129 mice from death and joint swelling against lethal Chikungunya virus challenge.

Li Jiangfan J, Fang Ting T, Song Liyuan L, Qiang Li L et al.

Chikungunya virus (CHIKV) is primarily transmitted to humans through bites of infected Aedes mosquitoes, causing febrile illness as well as acute or chronic arthritis. With increasing reports in previously non-endemic regions, it poses an emerging threat to public health and a growing risk to travelers. Despite the recent approval of live-attenuated and virus-like particle-based vaccines, a safe, efficacious, and cost-effective vaccine remains urgently needed. The envelope glycoproteins E1 and E2, which play crucial roles in viral attachment and entry, are the major immunodominant structural proteins and can effectively elicit protective antibodies. We engineered mammalian cells to produce the ectodomains of the E1 and E2 glycoproteins and evaluated the protective efficacy of the resulting vaccine formulations in vivo. A two-dose vaccination regimen elicited high levels of antibodies and provided full protection against lethal challenge in A129 mice, preventing both death and joint swelling. These findings demonstrate the potential of this vaccine candidate against CHIKV and warrant its further development.

Live and Heat-Killed Lactiplantibacillus plantarum HY7715 Attenuate D-Galactose-Induced Aging-Related Brain Changes and Blood-Brain Barrier Disruption with Gut Microbiota Modulation in Mice.

Baik Jimin J, Gwon Hyeonjun H, Kim Joo-Yun JY, Shim Jae-Jung JJ et al.

Aging is associated with changes in systemic homeostasis, intestinal barrier integrity, and brain function. This study investigated the effects of Lactiplantibacillus plantarum HY7715 on aging-related alterations in a D-galactose-induced accelerated aging mouse model. Male C57BL/6J mice were administered D-galactose for 8 weeks and orally treated with live or heat-killed HY7715. D-galactose increased circulating advanced glycation end products (AGEs), blood-brain barrier (BBB) permeability, hippocampal inflammation, and aging-related molecular changes, while impairing cortical and colonic tight junction-associated markers. Both live and heat-killed HY7715 reduced serum AGE levels, Evans blue leakage, and hippocampal inflammatory responses, while restoring CLDN5 immunoreactivity, tight junction-associated gene expression, and antioxidant-related gene expression. HY7715 also attenuated senescence- and apoptosis-related changes in the hippocampus and colonic tight junction- and inflammation-related gene expression changes. Gut microbiota analysis showed taxon-specific changes following HY7715 treatment, including significant increases in Bifidobacterium in both HY7715-treated groups, whereas live HY7715 was associated with significant changes in microbial community structure. Several microbial taxa were correlated with serum AGEs, BBB permeability, and neuroinflammation-related parameters. Together, these results show that both live and heat-killed HY7715 attenuated multiple aging-related changes in the D-galactose-induced accelerated aging model, accompanied by changes in gut microbiota composition.

Maternal Smoking and Sex Distribution Among Live Births in Western Sydney: A Population-Based Cohort Study.

Trinh Lieu Thi Thuy LTT, Clague Liesa L, Naz Sabrina S, Eljiz Kathy K

Maternal smoking may affect fetal survival, but evidence regarding offspring sex is inconsistent. We estimated the male proportion among live births and did not assess sex determination or pregnancy loss before the perinatal reporting threshold. To examine whether smoking during the first half of pregnancy was associated with male live birth and whether the association differed by smoking intensity. We analysed singleton live births of Western Sydney residents in the New South Wales Perinatal Data Collection, 2016-2023. Smoking was classified as non-smoking, ≤ 10 cigarettes/day, or > 10 cigarettes/day. Logistic regression estimated odds ratios (ORs), adjusted for calendar year and maternal demographic, socioeconomic and clinical characteristics. Of 119 756 eligible births, 721 (0.6%) had unknown smoking information, leaving 119,035 births. Male births comprised 51.6% among non-smokers, 52.2% among women smoking ≤ 10 cigarettes/day, and 52.4% among women smoking > 10 cigarettes/day. Crude differences were + 0.60 and + 0.78 percentage points (p = 0.608). The adjusted model included 117,745 births; adjusted ORs were 1.02 (95% CI 0.96-1.08) and 1.02 (95% CI 0.90-1.16), respectively. The heavier-smoking estimate was imprecise (n = 964). Lower-intensity smoking was not meaningfully associated with male live birth. Estimates for > 10 cigarettes/day were imprecise, and moderate effects in either direction could not be excluded. Findings apply only to live births reaching the reporting threshold.

The Senokine Gas6 Promotes Aortic Valve Calcification Through CaMKII-DRP1-Mediated Terminal Scission of Mitochondrial-Derived Vesicles.

Zhang Yaoyu Y, Zhang Jialiang J, Xu Yanjiani Y, Lei Wenhua W et al.

Calcific aortic valve disease (CAVD) is an age-associated valvular disorder in which calcification expands from discrete foci into adjacent tissue. We investigated whether mitochondrial-derived vesicles (MDVs) released by human valvular interstitial cells (hVICs) exert pro-calcific effects on recipient cells and how MDV release is regulated. Using ex vivo hemodynamic perfusion of human CAVD tissue, multi-omics profiling, live-cell single-organelle tracking, MDV transfer assays, and in vivo Gas6 deletion, we found that calcifying hVICs hypersecreted MDVs that exerted pro-calcific effects in recipient hVICs. High-resolution imaging showed that mitochondria remodeled into donut-shaped structures under pro-osteogenic conditions; these structures served as physical platforms for MDV budding. Human CAVD valve tissues and hVICs treated with pro-osteogenic medium also exhibited senescence-associated features. Multi-omics profiling identified growth arrest-specific 6 (Gas6), a senokine, among 36 concordantly upregulated candidates. Functional analyzes showed that Gas6 promoted terminal MDV scission while leaving donut-shaped mitochondrial precursor formation intact. Mechanistically, Gas6 promoted intracellular Ca2+ accumulation, calcium/calmodulin-dependent protein kinase II (CaMKII) activation, and dynamin-related protein 1 (DRP1) phosphorylation at Ser616, supporting a Ca2+-CaMKII-DRP1 axis that facilitates terminal MDV scission. Pharmacological inhibition of CaMKII or DRP1 reduced MDV hypersecretion and subsequent recipient-cell calcification, and Gas6 deletion in vivo attenuated valvular stenosis, calcification, and fibrotic remodeling. These findings identify Gas6-dependent terminal MDV scission as a mechanism contributing to the intercellular propagation of valvular calcification in a CAVD context characterized by senescence-associated features.

Clinical effects of third doses of mRNA COVID-19 vaccines among hemodialysis patients: a nationwide retrospective cohort study.

Cho AJin A, Jeong Seon A SA, Seo Yu Bin YB, Park Hayne Cho HC et al.

Evidence on the effectiveness of third-dose messenger RNA (mRNA) coronavirus disease 2019 (COVID-19) vaccination during the Omicron-dominant period in Asian hemodialysis (HD) patients remains limited. We evaluated the clinical effectiveness of a third mRNA vaccine dose against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection, hospitalization, and mortality in Korean HD patients. Using the Korea Disease Control and Prevention Agency-National Health Insurance Service database, we conducted a nationwide cohort study of HD patients aged ≥18 years who received two or three doses of BNT162b2 or mRNA-1273 before January 16, 2022. Study outcomes were SARS-CoV-2 infection, hospitalization, and death. Cox proportional hazards models were used to assess the associations between third-dose vaccination and clinical outcomes. Among 66,389 HD patients (mean age, 65.7 ± 12.9 years; 60.5% male), 55,125 (83.0%) received three vaccine doses and 11,264 (17.0%) received two doses. Receipt of three doses was associated with a significantly lower risk of SARS-CoV-2 infection (adjusted hazard ratio [aHR], 0.58; 95% confidence interval [CI], 0.56-0.60). Among 17,886 patients with SARS-CoV-2 infection, third-dose vaccine recipients had a lower risk of hospitalization (aHR, 0.65; 95% CI, 0.62-0.70) and 30-day mortality (aHR, 0.35; 95% CI, 0.30-0.41) compared with those who received two doses. Receipt of mRNA-1273 as the third dose was associated with lower observed risks of infection and hospitalization than receipt of BNT162b2. A third dose of an mRNA COVID-19 vaccine was associated with lower risks of SARS-CoV-2 infection, hospitalization, and mortality among HD patients during the Omicron-dominant period. These findings support the clinical importance of booster vaccination in this high-risk population.
