Comprehensive Stability Assessment of Squalene in a Nanoemulsion Adjuvant and the SpiN-Tec Vaccine: HPLC Quantification, Stress Testing, and Stability Studies.

Squalene-based nanoemulsions are widely used as adjuvants in vaccine formulations, but their stability can be affected by environmental factors such as pH, oxidative stress, temperature, and light. We have produced a squalene nanoemulsion (CTVad1) to support the clinical development of new vaccines. This study aimed to develop and validate an HPLC method for squalene quantification in SpiN-Tec, a recombinant protein vaccine against COVID-19. We also aimed to evaluate the stability of the CTVad1 adjuvant and SpiN-Tec under controlled storage conditions. A reversed-phase HPLC method was developed and validated, and comprehensive forced degradation studies were performed on the raw material and SpiN-Tec under acidic, basic, oxidative, thermal, and photolytic conditions to demonstrate the stability-indicating capability of the method. Physicochemical, morphological, and biological characteristics were assessed, and stability studies of both the vaccine and the CTVad1 adjuvant were performed under accelerated and long-term conditions. The HPLC method was selective, precise, accurate, linear, and robust. Squalene raw material degraded under all tested conditions, whereas formulation in nano-sized globules improved its stability, with degradation observed only under hydrogen peroxide and light exposure. CTVad1 remained stable over time, exhibiting only minor, non-critical changes within the specification limits in both accelerated and long-term stability studies, regardless of the glass packaging used (clear or amber). In addition, the SpiN-Tec vaccine maintained its physicochemical and biological integrity under all tested conditions, with all evaluated parameters remaining within the established specification ranges. Our findings demonstrate that proper formulation, packaging, and storage conditions can preserve squalene stability in nanoemulsion-based vaccines, ensuring the quality, safety, and efficacy of the SpiN-Tec vaccine and its adjuvant throughout shelf life.

Bilateral Maculopathy in an Infant Following Measles, Mumps, Rubella, and Varicella-Zoster (MMRV) Vaccination.

To report on the long-term clinical outcome of a healthy infant who presented with posterior uveitis following the administration of the combined measles, mumps, rubella, and varicella (MMRV) vaccine. Descriptive case report. A 13-month-old infant presented with bilateral visual loss two weeks after receiving the MMRV vaccine. Ophthalmic examination revealed bilateral retinitis and retinal vasculitis with exudative retinal detachment. Extensive infectious work-up was conducted including serologic exams and PCR testing of blood, aqueous humor, cerebrospinal fluid and urine. Serological tests for measles yielded positive IgM and IgG titers. PCR testing for measles in the aqueous humor and urine was negative. Neurologic assessment and neuroimaging were unremarkable, excluding central nervous system involvement. Empiric systemic antiviral and corticosteroid therapy was initiated. Gradual resolution of posterior uveitis ensued, culminating in bilateral macular scars. Over a six-year follow-up period, the patient demonstrated stable ocular findings with no evidence of recurrent inflammation or systemic autoimmune disease. Final visual acuity was 6/9 in each eye. This case represents a rare occurrence of non-necrotizing retinitis following MMRV vaccination. To our knowledge, this is the first report describing the sequential OCT characteristics of presumed measles vaccine-associated retinitis. The prolonged follow-up period supports the absence of an alternative etiology and provides valuable insight into the long-term course and visual prognosis of vaccine-associated retinopathy.

Measuring body checking and avoidance in adolescents: validation of a behavioral questionnaire.

Body image distortion (BID) is central to eating disorder (ED) pathology and is closely linked to body dissatisfaction. While most questionnaires assess affective, cognitive, or perceptual components of body image, the behavioral component manifesting in body checking (BC) and body avoidance (BA) has received comparatively little attention. Given that EDs typically emerge during adolescence, validated measures of behavioral BID are particularly needed for this age group. The Body Checking and Avoidance Questionnaire (BCAQ) is the only instrument assessing both BC and BA, but it has not yet been validated in adolescents. The present study aimed to evaluate the BCAQ in a clinical sample of female German adolescents with (atypical) anorexia nervosa. Factor structure, internal consistency, and convergent and known-groups validity were examined. An initial confirmatory factor analysis of the original 27-item BCAQ indicated poor model fit. An exploratory factor analysis identified four items with low or inconsistent loadings. Analyses of the resulting 23-item version yielded a preliminary three-factor structure and demonstrated good reliability (Cronbach's α = 0.78-0.94) and satisfactory convergent validity, while known-groups comparisons provided limited and exploratory evidence, with a small group difference observed only for avoidance behavior. These findings provide preliminary evidence for the use of the adapted 23-item BCAQ to assess behavioral aspects of body image distortion in adolescents with (atypical) anorexia nervosa. Further longitudinal research is needed to evaluate the measure's temporal stability.

Contextual and cultural influences on preschoolers' aesthetic experience: evidence from a situational teaching intervention in China.

Contextual and cultural influences play an important role in shaping young children's learning, meaning-making, and emotional engagement. In early childhood education, art appreciation provides a valuable context for examining how children perceive, interpret, and emotionally respond to culturally and pedagogically structured learning environments. However, empirical evidence remains limited regarding how culturally grounded contextual teaching approaches influence preschoolers' aesthetic experience. Drawing on situated learning theory and Li Jilin's situational teaching framework in China, this study examined the effects of a situational teaching intervention on preschoolers' aesthetic experience in art appreciation. A quasi-experimental pretest-posttest control group design was adopted. Sixty preschool children from two intact classes in a public kindergarten in Beijing, China, participated in the study. One class was assigned to the experimental group, and the other served as the control group. Over an eight-week period, the experimental group participated in situational art appreciation activities organized around life-related scenarios, storytelling, role-play, multisensory exploration, and reflective dialogue, whereas the control group received conventional art appreciation instruction. Children's aesthetic experience was assessed across three dimensions: aesthetic perception, aesthetic understanding, and aesthetic emotion. Repeated-measures ANOVAs revealed Group × Time interactions for visual perception and verbal expression, F(1, 58) = 8.12, p = 0.01, ηp 2 = 0.12; aesthetic understanding, F(1, 58) = 9.32, p < 0.001, ηp 2 = 0.14; facial expression, F(1, 58) = 5.14, p = 0.03, ηp 2 = 0.08; and preference explanation, F(1, 58) = 4.78, p = 0.03, ηp 2 = 0.08. No significant Group × Time interaction was found for viewing duration, F(1, 58) = 0.54, p = 0.46, ηp 2 = 0.01. After Bonferroni correction, robust intervention effects remained for visual perception and verbal expression and aesthetic understanding, while effects on aesthetic emotion were suggestive and should be interpreted with caution. The results suggest that culturally grounded situational teaching can support preschoolers' aesthetic-cognitive and socio-emotional engagement by embedding artworks within meaningful, interactive, and emotionally resonant learning contexts. This study provides educational psychological evidence for the role of contextualized teaching in promoting young children's perception, meaning-making, and emotional expression. It also extends research on contextual and cultural influences by showing how a Chinese situational teaching model may function as a developmentally appropriate contextual learning approach in early childhood art education.

Lifestyle clusters and bone mineral density in Chinese adults: a cross-sectional study with cluster analysis.

Osteoporosis is a major global public health challenge, with lifestyle as a key modifiable factor. This study aimed to examine associations between lifestyle patterns and bone mineral density (BMD) in Chinese adults. In this cross-sectional, multi-center study, 5,324 adults from Beijing (2016-2020) with complete lifestyle questionnaires were analyzed (lumbar spine BMD available for 4,028; hip BMD available for 4,654). Each of eight lifestyle variables was residualized on age and sex before k-means clustering (k = 3) categorized participants into three groups: office/mental-labor workers with low leisure-time activity (OL), physical-labor workers (PL), and office/mental-labor workers with an active leisure-time lifestyle (OA). Differences in lumbar spine and hip BMD among clusters were examined using multivariable-adjusted models (age modeled via restricted cubic spline, sex, body mass index [BMI], hospital fixed effects), with stratification by sex, age group, and BMI group, alongside full-cohort dose-response and cluster × exposure interaction analyses. After adjustment, the OA cluster showed higher lumbar spine BMD than the OL cluster (adjusted difference = 0.024 g/cm2, 95% CI 0.012-0.036, P < 0.001). The PL cluster showed lower hip BMD than both the OL cluster (adjusted difference = -0.041 g/cm2, 95% CI -0.056 to -0.027, P < 0.001) and the OA cluster (adjusted difference = 0.028 g/cm2, 95% CI 0.011-0.044, P = 0.002), despite the occupational physical activity inherent to manual labor. These associations were broadly consistent across sex, age, and BMI strata, though attenuated among participants aged ≥70 years and those with obesity. In dose-response analyses, exercise and sun exposure showed non-linear associations with BMD at both sites, while calcium supplementation frequency showed a consistent negative linear association that did not vary by cluster. A lifestyle pattern combining active leisure-time behavior with office-based occupation was associated with higher lumbar spine BMD, while an occupationally physical-labor lifestyle was associated with lower hip BMD relative to both other clusters. These associations exhibited site-specific patterns and were generally consistent across demographic subgroups. Given the cross-sectional design, these findings should be interpreted as associations rather than causal effects; prospective studies are needed to clarify their causal structure and clinical significance.

Neoantigen cancer vaccines for gastrointestinal tumors: opportunities and challenges.

Gastrointestinal tumors are characterized by high global incidence and mortality rates. Although immune checkpoint inhibitors (ICIs) have achieved breakthroughs in some of these cancers, their overall response rate remains low. Neoantigen cancer vaccines have emerged as a promising new strategy for immunotherapy in gastrointestinal tumors due to their high specificity and strong immunogenicity. These vaccines effectively activate specific T-cell immunity and can produce synergistic effects when combined with ICIs. Various vaccine platforms offer distinct advantages, and clinical trials have shown encouraging potential in inducing immune responses and extending progression-free survival. Meanwhile, current challenges and future directions cannot be ignored. Key challenges primarily involve the accuracy of neoantigen prediction, tumor heterogeneity, and optimal treatment timing. Future directions include artificial intelligence (AI)-assisted multi-omics screening, the development of universal vaccines, optimization of novel delivery systems, and multimodal combination strategies. These advances are expected to promote the application of neoantigen cancer vaccines in postoperative recurrence prevention and early-stage treatment, ultimately moving toward personalized precision immunotherapy. This article systematically reviews the clinical progress and prospects of neoantigen cancer vaccines in treating gastrointestinal tumors, aiming to provide insights for immunotherapy in this field and offer new perspectives for further optimization of such vaccines.
