June 8, 2026: Tango Therapeutics, Inc. (NASDAQ: TNGX) reported positive initial data from an ongoing Phase 1/2 study of the PRMT5 inhibitor vopimetostat combined with Revolution Medicines’ RAS(ON) inhibitors daraxonrasib or zoldonrasib in previously treated MTAP-deleted, RAS-mutant metastatic pancreatic ductal adenocarcinoma (PDAC) (and a small NSCLC cohort). Data cutoff was May 28, 2026.
Vopimetostat + daraxonrasib (PDAC/NSCLC dose escalation): In response-evaluable PDAC patients (n=12), ORR was 92% (11/12; 9 confirmed), 6-month PFS rate 90% (median PFS not reached), and DCR 100%. In NSCLC (n=3), ORR was 100% (3/3 confirmed). Generally well tolerated; no related grade 4/5 AEs, no DLTs at 200 mg vopimetostat/100 mg daraxonrasib, and 3 DLTs in two patients at the higher vopimetostat dose.
Vopimetostat + zoldonrasib (PDAC with MTAP deletion and KRAS G12D): In response-evaluable patients (n=27), ORR was 52% (14/27; 10 confirmed), 6-month PFS rate 74%, DCR 96%; generally well tolerated with no DLTs.
Tango plans (subject to regulatory feedback) rapid advancement of vopimetostat + daraxonrasib into Phase 3 in first-line MTAP-deleted PDAC; Phase 3 design finalization targeted for 2H 2026. Other 2H 2026 milestones include vopimetostat lung cancer monotherapy data, initial TNG456 glioblastoma data, a scientific conference presentation of the 2L/3L PDAC combination data, and initiation of a vopimetostat + ERAS-0015 Phase 1/2 study.
