Optune (EF-32) - (TRIDENT)

June 18, 2026: Novocure (NASDAQ: NVCR) reported topline results from the Phase 3 TRIDENT trial in newly diagnosed glioblastoma, comparing Tumor Treating Fields (TTFields) initiated at the start of chemoradiation (Early Start) vs initiation during the maintenance phase (Maintenance Start). TRIDENT did not meet its primary endpoint of a statistically significant overall survival (OS) improvement: median OS was 17.7 months in the Early Start arm vs 17.5 months in the Maintenance Start arm (HR 0.953; p=0.519) in the intent-to-treat population. One-, two-, and three-year survival rates were 70.9%, 33.9%, and 22.5% (Early Start) vs 72.0%, 31.6%, and 18.4% (Maintenance Start). Safety was consistent with prior TTFields GBM studies, with no new safety signals, and early initiation during chemoradiation was feasible and well-tolerated.

TRIDENT enrolled 981 patients randomized shortly after surgery; ~25% did not initiate the maintenance phase. Results have been accepted for presentation at the ASTRO 2026 Annual Meeting, and the company plans additional analyses.
