March 9, 2026: Actuate Therapeutics, Inc. (NASDAQ: ACTU) announced an expanded preclinical research initiative to evaluate combinations of its clinical-stage GSK-3β inhibitor elraglusib with emerging RAS-targeted therapies for RAS-driven cancers. The company cited adaptive resistance and pathway reactivation as key challenges with RAS inhibitors and described a mechanistic rationale for adding GSK-3β inhibition to suppress downstream survival and resistance pathways (including NF-κB signaling, MYC-driven programs, metabolic adaptation/oxidative stress buffering, and immune suppression). Actuate also highlighted potential immune-modulating effects of GSK-3β inhibition in RAS-mutant tumors (notably pancreatic cancer).
Elraglusib has been administered to more than 500 patients and is currently being evaluated in a Phase 2 trial in metastatic pancreatic cancer.
Catalyst/timing: Initial preclinical combination data are expected in Q2 2026, with additional results expected in 2H 2026. The planned work includes in vitro studies in RAS-resistance models, in vivo tumor regression/survival studies, and translational biomarker analyses, with involvement of academic collaborators and potential industry partners.
