ALLO-329 - (RESOLUTION)

April 15, 2026: Allogene Therapeutics, Inc. (NASDAQ: ALLO) announced publication of preclinical data for ALLO-329, an investigational allogeneic CD19/CD70 dual CAR T product for autoimmune diseases, in Nature Communications. The paper describes ALLO-329’s targeting of CD19+ B cells and CD70+ activated T cells; in preclinical models, adding an anti-rejection CD70 CAR to allogeneic CD19 CAR T cells preserved anti-CD19 activity, reduced rejection, and supported CAR T expansion/persistence. The dual-targeting approach also showed improved efficacy in tumor models of antigen loss and suppressed antibody titers in an autoimmune disease model; dual CD19/CD70 CAR T cells rapidly eliminated B and T cells and halted autoantibody production in systemic lupus erythematosus (SLE) models. The company highlighted its Dagger® technology as enabling expansion/persistence with the aim of reducing or eliminating cytotoxic lymphodepletion.

Clinical catalyst: the Phase 1 RESOLUTION 3+3 dose-escalation “rheumatology basket” trial is actively enrolling SLE, scleroderma, and inflammatory myositis, testing doses starting at 20 million CAR T cells in cohorts with cyclophosphamide-only reduced lymphodepletion vs no lymphodepletion. Initial first-dose-level data are expected June 2026; an additional clinical update is planned for year-end 2026.
