November 6, 2026: MiNK Therapeutics (NASDAQ: INKT) reported two preclinical SITC 2026 posters and highlighted ongoing Phase 2 programs for its iNKT cell platform.

agenT-797 / MSS colorectal cancer liver metastases: In a human organoid model of checkpoint inhibitor-refractory colorectal cancer liver metastases, agenT-797 shifted the microenvironment toward immune stimulation, reducing CD206+ M2-like macrophages and increasing CD86+/HLA-DR+ antigen-presenting cells. Combined with antigen-specific T cells plus botensilimab and balstilimab, it increased CD8 T-cell density, granzyme B, IFN-γ, TNF-α, CXCL-9, CCL3 and IL-8, reduced EpCAM+ tumor cells, and increased apoptosis. BOT+BAL alone showed no statistically significant tumor-cell killing. MiNK said this supports its ongoing Phase 2 combination study in previously treated MSS CRC with liver metastases (NCT07550088).

agenT-797 and MiNK-215 / senescence models: In collaboration with Imperial College London, both unmodified agenT-797 and FAP-targeted, IL-15-armored CAR-iNKT candidate MiNK-215 killed therapy-induced senescent breast cancer cells. MiNK-215 also killed FAP-high lung fibroblasts; senescent lung fibroblasts resisted both therapies and showed elevated PD-L1, PD-L2, and CD155. α-GalCer restored unmodified agenT-797 killing in less-sensitive senescent fibroblasts.

Pulmonary program: MiNK said these findings support further investigation in its randomized, placebo-controlled Phase 2 trial of agenT-797 in acute hypoxemic respiratory failure/ARDS (C-1300-02; NCT07615010).
