Selinexor (XPORT-MF-034) - (SENTRY)

Nov. 4, 2026: Karyopharm Therapeutics (NASDAQ: KPTI) said three selinexor abstracts were accepted for presentation at ASH 2026.

Myelofibrosis — Phase 3 SENTRY (XPORT-MF-034; NCT04562389): week-48 data on selinexor plus ruxolitinib in JAK inhibitor-naïve myelofibrosis will be presented orally on Dec. 13, 2026 (Abstract 526). The randomized Phase 3 trial compares once-weekly selinexor 60 mg plus ruxolitinib versus placebo plus ruxolitinib in 353 patients with platelet counts >100 x 10^9/L; co-primary endpoints are SVR35 at week 24 and average change in absolute total symptom score over 24 weeks. The company said ongoing benefit was observed through week 48.

Myelofibrosis — Phase 2 SENTRY-2 (XPORT-MF-044; NCT05980806): oral presentation on Dec. 12, 2026 (Abstract 244) will cover selinexor monotherapy in JAKi-naïve myelofibrosis with thrombocytopenia. The company said early 60 mg cohort data support monotherapy activity. The primary endpoint is week-24 SVR35; optional add-on ruxolitinib, pacritinib, or momelotinib is allowed for inadequate spleen response.

Multiple myeloma — STOMP Arm 12 (NCT02343042): updated poster data on selinexor, mezigdomide, and dexamethasone in triple-class exposed relapsed/refractory multiple myeloma will be presented Dec. 12, 2026 (Abstract 2242).
