September 24, 2026: Merck (NYSE: MRK) reported topline 52-week results from the pivotal Phase 2b/3 BRUNELLO trial (NCT06571045) of remigromig (MK-3000, formerly EYE103) in adults with diabetic macular edema. Both remigromig doses (0.5 mg and 0.8 mg) independently demonstrated non-inferiority to 0.5 mg ranibizumab for mean change from baseline in best-corrected visual acuity at week 52. Remigromig was generally well tolerated, but higher rates of proliferative diabetic retinopathy, vitreous hemorrhage, and adverse-event discontinuations were reported versus ranibizumab; Merck said further analyses are underway and the results will be discussed with regulators.

BRUNELLO enrolled 984 participants randomized 1:1:1 to low-dose remigromig, high-dose remigromig, or ranibizumab every four weeks in year 1; year 2 shifts to a personalized treatment interval algorithm.

Upcoming catalyst: BRUNELLO 1-year results will be presented at the American Academy of Ophthalmology Annual Meeting in New Orleans on October 10 at 5:10 p.m. CT.

Pipeline updates mentioned: remigromig is also in the ongoing pivotal Phase 2b/3 BAROLO study in DME (NCT06957080) and a Phase 2 proof-of-concept study, SUPER TUSCAN, in neovascular AMD and retinal vein occlusion (NCT07205887). Merck also highlighted MK-8748/Tiespectus (EYE201), in pivotal Phase 2b/3 NVAMD trials TORRONTES and MALBEC and Phase 3 DME trials SANGIOVESE and SYRAH.
