September 28, 2026: Arvinas, Inc. (NASDAQ: ARVN) announced the first patient was dosed in ARV-6723-101 (NCT07749586), a global Phase 1/2 first-in-human trial of ARV-6723, an oral PROTAC HPK1 degrader, in adults with advanced solid tumors. The study will evaluate ARV-6723 monotherapy and later ARV-6723 plus pembrolizumab, assessing safety, pharmacokinetics, pharmacodynamics, and preliminary antitumor activity. The initial monotherapy cohort is enrolling patients previously treated with an immune checkpoint inhibitor who have no suitable standard treatment options.
The catalyst is a clinical trial initiation/first-patient-dosed milestone for Arvinas’ first immuno-oncology candidate and, according to the release, the first HPK1 PROTAC degrader to enter the clinic in the U.S.
Supporting preclinical data cited in the release showed potent, selective HPK1 degradation, enhanced immune activity, and antitumor activity as both a single agent and in combination with an immune checkpoint inhibitor across tumor models with differing immune responsiveness, including multiple anti-PD-1-resistant models. In seven preclinical models, ARV-6723 showed meaningful single-agent activity where neither an HPK1 inhibitor nor anti-PD-1 therapy showed benefit.
