Sept. 28, 2026: ALX Oncology (NASDAQ: ALXO) announced a Nature Medicine publication of Phase 2 ASPEN-06 data for evorpacept in HER2-positive advanced gastric/gastroesophageal junction cancer. In previously treated second-/third-line patients, evorpacept plus trastuzumab, ramucirumab and paclitaxel achieved a 40.3% ORR versus 26.6% for control in the intent-to-treat population. In patients with retained HER2-positive disease, ORR was 48.9% versus 25.0%; in the post-hoc subgroup with retained HER2 positivity and high CD47 expression, ORR was 63.6% versus 23.1%, median PFS 19.5 vs 7.0 months, and median DOR 25.5 vs 8.4 months. The company said these analyses support CD47 expression as a predictive biomarker.

ALX said the data suggest meaningful benefit with manageable safety, but it will not pursue a U.S. registrational Phase 3 path in gastric cancer and may seek development partnerships for that program.

The company said ASPEN-06 supports the ongoing Phase 2 ASPEN-09-Breast trial of evorpacept plus trastuzumab and chemotherapy in HER2-positive metastatic breast cancer, with topline data from 80 patients expected in mid-2027.
