Sept. 28, 2026: Silexion Therapeutics Corp. (NASDAQ: SLXN) reported positive translational research findings for SIL204, its lead siRNA candidate in Phase 2/3 development for locally advanced pancreatic cancer. In a carrier-free, non-transfection assay, human KRAS-driven cancer cells showed robust, dose-dependent mutant KRAS knockdown with SIL204 in the presence of normal physiological serum lipids. Removing serum lipids significantly reduced gene-silencing efficiency (P < 0.001), supporting a direct role for circulating lipoproteins in cellular uptake. Adding exogenous human LDL did not improve potency versus complete serum alone, indicating native lipoprotein levels are sufficient for delivery without artificial nanoparticles or external lipid supplementation.

The company said these findings support the mechanistic rationale for systemic subcutaneous administration and its broader clinical development strategy. Silexion also said translational characterization of SIL204 is continuing alongside clinical trial site initiation at Tel Aviv Sourasky Medical Center and regulatory clearances in Germany. Additional preclinical studies are underway, including in vivo pharmacokinetic, cellular uptake kinetics, and tissue biodistribution evaluations.
