Sept. 25, 2026: Johnson & Johnson (NYSE: JNJ) reported long-term follow-up from cohort A of the Phase 2 CARTITUDE-2 study of CARVYKTI (ciltacabtagene autoleucel; cilta-cel) in early-line relapsed/refractory multiple myeloma and presented the data at the 2026 International Myeloma Society Annual Meeting (Abstract PA-288).
In 20 patients with RRMM who had received 1-3 prior lines of therapy, prior proteasome inhibitor exposure, and lenalidomide-refractory disease, a single CARVYKTI infusion without maintenance therapy produced 5-year progression-free survival in 50% (10/20). At a median follow-up of 60.7 months, the 5-year overall survival rate was 69.2% and median progression-free survival was 60.5 months. Among three patients assessed for MRD at five years, all were MRD-negative at 10^-6. Durable remissions were also seen in high-risk patients; 5 of the 10 patients alive and progression-free at five years had at least one high-risk cytogenetic abnormality.
Safety was consistent with the known profile, with no new CAR T-cell-related neurotoxicity reported. Since the previous analysis, one patient developed acute myeloid leukemia and two deaths occurred from progressive disease and new cancer.
