NHBE GalNAc siRNA (WVE-007) - (INLIGHT)

Sept. 23, 2026: Wave Life Sciences (NASDAQ: WVE) announced initiation of a Phase 2a trial of WVE-007, an investigational INHBE GalNAc-siRNA, in combination with weekly tirzepatide in adults with obesity and type 2 diabetes. The placebo-controlled 3:1 study will test 240 mg and 400 mg doses in patients with BMI 35-50 kg/m2.

WVE-007 is being developed in the INLIGHT program across three obesity settings: monotherapy, combination therapy, and post-incretin maintenance. Wave said prior clinical data support fat loss without muscle loss and potential once- or twice-yearly dosing, while preclinical data previously presented at ObesityWeek 2025 showed WVE-007 combined with GLP-1 therapy doubled weight loss versus GLP-1 alone and may help limit weight regain after GLP-1 discontinuation.

The company also said its ongoing Phase 2a monotherapy trial includes multiple assessments over 12 months, with results expected to inform further development in obesity as well as MASH, type 2 diabetes, and cardiovascular disease. A separate maintenance trial evaluating WVE-007 after incretin cessation remains on track to start in 2H 2026.
