Sept. 10, 2026: BioLineRx Ltd. (NASDAQ/TASE: BLRX) and Hemispherian AS said two GLIX1 abstracts were accepted for EANO 2026 (Sept. 24-27, Rome), including a mini-oral presentation on preclinical glioblastoma data and an e-poster on the ongoing first-in-human Phase 1/2a trial design.
GLIX1 is an oral TET2 activator targeting the DNA damage response. In preclinical work, it showed high in vitro potency, increased TET2-dependent 5hmC generation, and brain exposure in mice of 68-85% of plasma levels, supporting CNS penetration. In two orthotopic GBM xenograft models (SNB19 and U87-MG), oral GLIX1 at 75 mg/kg QD to 1000 mg/kg BID produced dose-dependent tumor growth inhibition and survival benefit at all tested doses, with activity seen even at the lowest dose; temozolomide was the positive control. The companies said these findings supported initiation of the Phase 1/2a study.
The ongoing open-label, multicenter Phase 1/2a study (NCT07464925) is enrolling recurrent or progressive high-grade glioma patients. Using a BOIN dose-escalation design, it will test up to 5 dose levels in up to 30 patients. Primary endpoints are safety, tolerability, and MTD/RP2D; secondary endpoints include PK, antitumor response, and 6-month PFS. Preclinical and initial clinical data will be presented at EANO.
