ADI-212

August 27, 2026: Adicet Bio, Inc. (NASDAQ: ACET) said the FDA cleared its IND application for ADI-212 in metastatic castration-resistant prostate cancer (mCRPC). The company plans to begin Phase 1 enrollment in mCRPC in the fourth quarter of 2026.

ADI-212 is described as a next-generation, gene-edited allogeneic gamma delta CAR T-cell therapy targeting PSMA. The program includes a novel CAR binder intended to improve tolerability and tumor-specific recognition, membrane-tethered IL-12 armoring, and CRISPR/Cas9-mediated disruption of MED12, with the goal of improving potency in solid tumors and supporting multiple anti-tumor mechanisms in the tumor microenvironment.

The announcement is a regulatory milestone and start-of-clinic update; no clinical efficacy or safety data were reported in the release.
