vopimetostat (TNG462) in combination with RAS(ON) inhibitors (RMC-9805)

August 11, 2026: Tango Therapeutics (NASDAQ: TNGX) reported Q2 2026 updates highlighting vopimetostat, its MTAP-selective once-daily PRMT5 inhibitor. Initial Phase 1/2 data in MTAP-deleted, RAS-mutant pancreatic cancer showed vopimetostat plus Revolution Medicines’ RAS(ON) inhibitor daraxonrasib achieved a 92% objective response rate and 90% six-month progression-free survival, with a generally well-tolerated safety profile. Based on these results, Tango said it is working with regulators and collaborator Revolution Medicines on a registrational path for vopimetostat plus daraxonrasib in MTAP-deleted pancreatic cancer.

Upcoming catalysts for vopimetostat: Phase 1/2 combination data with RAS(ON) inhibitors will be presented at the 2026 ESMO Congress in Madrid on October 23-27, 2026; the company expects to finalize the design of a Phase 3 randomized-controlled trial in front-line pancreatic cancer in 2H 2026; vopimetostat lung cancer monotherapy data are expected in 2H 2026; and a Phase 1/2 study of vopimetostat plus ERAS-0015 is planned to start in 2H 2026.

Tango also announced Fatma Ocak as Chief Commercialization Officer, effective August 17, 2026, and said Robert Azelby was appointed Chairman on August 6, 2026. Cash, cash equivalents, and marketable securities were $1.0 billion as of June 30, 2026.
