August 11, 2026: Shattuck Labs (NASDAQ: STTK) reported Phase 1 results and upcoming milestones for its DR3 programs. For SL-325, a DR3-blocking antibody for Crohn’s disease, ulcerative colitis, and other inflammatory/immune-mediated diseases, Phase 1 showed a favorable safety profile, no evidence of DR3 agonism, 3.7% antidrug antibody incidence, and complete TL1A-to-DR3 blockade for over 3 months at doses of 1 mg/kg and higher, supporting potential quarterly maintenance dosing.
Shattuck said the randomized, double-blind, placebo-controlled RECEPTIVE-CD1 Phase 2b trial in moderately-to-severely active Crohn’s disease is expected to start in Q3 2026. The study will enroll about 232 patients randomized 1:1:1:1 across three SL-325 dose levels and placebo. The primary endpoint is endoscopic response at 12 weeks, with data expected in 1H 2028; treatment continues through a 50-week maintenance phase with long-term extension eligibility.
For SL-846, a half-life extended DR3 x IL-23 receptor bispecific antibody, the program is in an IND-enabling GLP non-human primate toxicology study. Preclinical safety, PK, and receptor occupancy data are expected in a poster at UEG Week on October 17-20, 2026. A Phase 1 healthy volunteer trial is planned, with initial data expected in 2027.
Shattuck reported $208.3 million in cash/investments as of June 30, 2026, expected to fund operations into 2029.
