August 6, 2026: Structure Therapeutics (NASDAQ: GPCR) said first patients have been dosed in its Phase 3 ACCOMPLISH program for aleniglipron, an oral small-molecule GLP-1 receptor agonist for chronic weight management. ACCOMPLISH-1 will enroll up to 3,600 adults with obesity or overweight plus a weight-related comorbidity; ACCOMPLISH-2 will enroll up to 1,100 adults with obesity or overweight and type 2 diabetes. Both are randomized, double-blind, placebo-controlled trials testing 45 mg, 90 mg, and 180 mg maintenance doses versus placebo, intended to support global regulatory submissions.

For aleniglipron, the company highlighted June 2026 Phase 2b ACCESS data published in Nature Medicine and presented at ADA, showing dose-dependent, statistically significant weight loss, continued loss beyond 36 weeks, up to 16.2% at 44 weeks in the open-label extension, and a 10.4% overall discontinuation rate. Upcoming aleniglipron catalysts: 72-week ACCESS OLE data in Q3 2026; body composition, type 2 diabetes/obesity, and SWITCH trial data in Q4 2026.

ACCG-2671, an oral dual amylin/calcitonin receptor agonist, remains on track for initial Phase 1 SAD data and MAD trial initiation in Q3 2026. ACCG-3535 Phase 1 initiation and an oral GLP-1/amylin combination trial are expected in Q4 2026. Cash was $1.3 billion as of June 30, 2026, with runway through 2028.
