July 30, 2026: Evommune, Inc. (NYSE: EVMN) announced dosing of the first patient in a global Phase 2b, randomized, double-blind, placebo-controlled, dose-ranging trial of EVO756, an oral, highly selective MRGPRX2 antagonist, for prophylactic treatment of refractory migraine. The study will enroll ~330 adults with ≥6 migraine days per month, randomized to two active dose arms or placebo for 12 weeks; daily doses up to 100 mg are being evaluated.

Primary endpoint is mean change from baseline in monthly migraine days (MMD). Key secondary endpoints include ≥50% and ≥75% MMD responder rates, change in monthly headache days, and acute migraine medication use; exploratory endpoints include patient subtyping, migraine-related biomarkers, and quality-of-life measures.

The company highlighted MRGPRX2 expression in human trigeminal neurons and meningeal mast cells and said EVO756 is designed to inhibit three neuropeptides (PACAP, VIP, substance P) implicated in triggering migraine signaling through MRGPRX2. Top-line data are expected in 2027.
