VS-7375 -(TARGET-D 203)

July 28, 2026: Verastem Oncology (NASDAQ: VSTM) announced first-patient dosing in TARGET-D 203 (NCT07659795), a Phase 2, open-label, registration-directed trial of VS-7375 (oral KRAS G12D ON/OFF inhibitor) in KRAS G12D-mutated metastatic colorectal cancer (mCRC). TARGET-D 203 evaluates VS-7375 900 mg QD as monotherapy and in combination with anti-EGFR therapies (cetuximab or panitumumab) in previously treated mCRC, and also includes a cohort combining VS-7375 with cetuximab plus chemotherapy in the first-line setting.

The company said treatment has now been initiated across all three TARGET-D Phase 2 registration-directed trials in KRAS G12D-mutated cancers: TARGET-D 201 in second-line advanced/metastatic pancreatic ductal adenocarcinoma (first patient dosed June 2026) and TARGET-D 202 in second/third-line advanced/metastatic NSCLC (first patient dosed July 2026), in addition to TARGET-D 203.

Verastem noted preliminary activity and no unexpected toxicities for VS-7375 plus full-dose cetuximab from its ongoing TARGET-D 101 Phase 1/2 study (initiated June 2025) and expects a further update in 2H 2026. FDA Fast Track Designations were granted in July 2025 (pancreatic cancer settings) and June 2026 (post-platinum/post–PD-(L)1 NSCLC).
