Retatrutide - (TRIUMPH)

July 23, 2026: Eli Lilly and Company (NYSE: LLY) announced positive topline results from two pivotal Phase 3 trials of retatrutide, an investigational once-weekly GIP/GLP-1/glucagon triple receptor agonist, in obesity populations with serious complications.

TRIUMPH-2 (NCT05929079; 80 weeks; n=1,152) in adults with obesity or overweight and type 2 diabetes: retatrutide met the primary endpoint (percent change in body weight at 80 weeks). Mean weight loss was 12.7% (29.8 lbs) with 4 mg, 19.1% (45.4 lbs) with 9 mg, and 20.8% (49.6 lbs) with 12 mg vs 4.0% (9.3 lbs) with placebo. A1C decreased by up to 1.6% (vs 0.2% placebo).

TRIUMPH-3 (NCT05882045; 80 weeks; n=1,949) in severe obesity (BMI ≥35) with established cardiovascular disease (with/without type 2 diabetes): retatrutide met the primary endpoint with mean weight loss of 21.6% (52.7 lbs) at 9 mg and 22.6% (55.8 lbs) at 12 mg vs 3.2% (7.7 lbs) placebo. Pre-specified MACE-5 HR was 0.82 (95% CI 0.55–1.22) and MACE-3 HR 1.12 (95% CI 0.64–1.96). Risk factors (e.g., triglycerides, non-HDL, SBP, hsCRP) improved.

Lilly said detailed data will be presented later and plans to submit a BLA to FDA for retatrutide in Q1 2027.
