Daraxonrasib (RMC-GI-102) plus gemcitabine and nab-paclitaxel - (RASolute 303)

July 2, 2026: Revolution Medicines, Inc. (NASDAQ: RVMD) reported Phase 1/2 results for zoldonrasib (RMC-9805), an oral RAS(ON) G12D-selective inhibitor, in combination regimens for RAS G12D metastatic pancreatic ductal adenocarcinoma (PDAC), to be presented in a proffered paper session at the 2026 ESMO Gastrointestinal Cancers Congress.

Zoldonrasib + chemotherapy (RMC-GI-102; NCT06445062; data cutoff Feb 8, 2026): In previously untreated metastatic RAS G12D PDAC, 41 patients received zoldonrasib + modified FOLFIRINOX (mFFX) and 40 received zoldonrasib + gemcitabine/nab-paclitaxel (GnP). Grade ≥3 TRAEs occurred in 61% (mFFX) and 80% (GnP); no Grade 5 TRAEs. ORR/DCR were 82%/96% (mFFX) and 61%/90% (GnP). These data support the ongoing Phase 3 RASolute 305 trial (NCT07621718) of zoldonrasib + chemotherapy vs placebo + chemotherapy in first-line metastatic RAS G12D PDAC.

Zoldonrasib + daraxonrasib (RMC-9805-001; NCT06040541; data cutoff Feb 9, 2026): In 60 previously treated RAS G12D metastatic PDAC patients, Grade ≥3 TRAEs were 35% (notably rash 12%, anemia 10%). ORR was 50% (2L; n=30) and 47% (3L+; n=30); median PFS 9.6 months (2L) and 7.6 months (3L+). These results support a planned Phase 3 RASolute 309 trial comparing the doublet vs GnP in previously untreated RAS G12D metastatic PDAC.
