June 30, 2026: ADC Therapeutics SA (NYSE: ADCT) announced completion of enrollment in LOTIS-7, a Phase 1b open-label trial evaluating ZYNLONTA (loncastuximab tesirine-lpyl) plus the bispecific antibody glofitamab (COLUMVI) in relapsed/refractory diffuse large B-cell lymphoma (r/r DLBCL). A total of 100 r/r DLBCL patients were dosed at the selected ZYNLONTA 150 µg/kg dose in the glofitamab arm across 30 sites (70% US/30% EU); baseline characteristics included 46% relapsed and 54% primary refractory, median age 66.
The primary endpoints are safety/tolerability; secondary endpoints include ORR, DoR, CR, relapse-free survival, PFS, OS, plus PK/immunogenicity. The company referenced previously reported results in 49 efficacy-evaluable patients (≥6 months follow-up) showing 89.8% ORR, 77.6% CR, and a manageable safety profile.
Next catalysts: full LOTIS-7 data are expected in 4Q 2026, with presentation at a medical meeting and submission for publication by end of 2026; the company also plans to assess potential regulatory and compendia pathways for the combination.
