PIPE-791

June 29, 2026: Contineum Therapeutics, Inc. (NASDAQ: CTNM) announced publication in the Journal of Medicinal Chemistry (Chen et al.) describing the discovery and characterization of PIPE-791, a selective lysophosphatidic acid receptor 1 (LPAR1) antagonist with slow, tight-binding properties and brain penetration. The manuscript reports that structural-activity work identified a scaffold linked to slow on/off binding to LPAR1, and subsequent optimization improved blood-brain barrier penetration and ADME, enabling once-daily oral dosing with full receptor occupancy. Preclinical disease models are cited as supporting potential use in CNS and peripheral disorders driven by LPA–LPAR1 signaling, including fibrotic conditions.

Catalyst/status: PIPE-791 is currently in a global Phase 2 clinical trial in idiopathic pulmonary fibrosis (IPF) (ClinicalTrials.gov NCT07284459). The release also notes PIPE-791 is in development for IPF and chronic pain, and references PIPE-307 (M1 receptor inhibitor) in clinical development for relapsing-remitting multiple sclerosis and major depressive disorder, with development under a license agreement with Janssen Pharmaceutica NV.
