June 29, 2026: Alaunos Therapeutics, Inc. (NASDAQ: TCRT) reported a new post hoc statistical analysis from its previously disclosed non-GLP 48-day diet-induced obesity (high-fat diet) mouse Study 1 of ALN1003, an investigational oral, non-hormonal, non-incretin small-molecule metabolic candidate. In an ANCOVA adjusting liver weight for body fat percentage, ALN1003-treated animals had lower liver weight vs controls (p=0.000005), confirmed with HC3 robust standard errors (p=0.000015), suggesting the liver-weight effect was not fully explained by measured body fat in this model.
The company said the adjusted liver-weight result is directionally consistent with earlier disclosed Study 1 findings, including: a blinded pilot pathology review of selected livers suggesting lower hepatic steatosis and lower mean NAFLD Activity Score (2.7 vs 5.0 in selected controls), a reported 43% reduction in absolute liver weight, and lower liver injury enzymes (ALT, AST, ALP); lower fasting insulin and lower HOMA-IR, with body-fat-adjusted HOMA-IR remaining significant (ANCOVA p=0.0006; HC3 p=0.0014); and adipose endocrine biomarker changes (numerically lower leptin, higher adiponectin, higher adiponectin-to-leptin ratio). Alaunos emphasized these are limited, non-GLP mouse data with multiple stated limitations; ALN1003 has not been tested in humans.
